During my PhD, I have generated a spontaneous high-grade glioma mouse model that is useful for testing novel treatment strategies against brain cancer. Currently, I am investigating the role of polycomb group protein Ezh2 on tumor initiation and tumor maintenance in established mouse models for Glioblastoma Multiforme (GBM) and human GBM cell lines. Using an inducible shRNA approach we identified several enriched genes upon Ezh2 knockdownin vivowhich are involved in DNA damage response and pluripotency. Next, we will explore combined therapies of Ezh2 inhibition, alkylating agents and molecular targeted drugs, such as DNA repair inhibitors.
